Short answer: Target tablet weight equals your active ingredient's dose divided by its percentage of the total formula — so a 200 mg active dosed at 40% of the tablet gives a target weight of 500 mg (200 ÷ 0.40). Once you know the target weight, convert it to a fill depth by running a short calibration press: weigh 10–20 tablets at a trial fill-depth setting, find the average weight, and adjust the depth up or down until the average lands on target. Confirm it holds with a weight-variation check across a full test batch before locking in the setting for a production run.
This is arguably the single most important calculation in tableting — get it wrong and every tablet either under- or over-doses, regardless of how well the press itself is running. The good news is it only takes two steps: figure out the weight on paper, then translate that weight into a physical press setting.
Step 1: Calculate Target Tablet Weight
The formula is simple once you know your active ingredient's dose and what percentage of the total tablet it's supposed to make up:
Target tablet weight = active ingredient dose ÷ active ingredient's percentage of the formula
For example: if your formula is built to deliver 200 mg of active ingredient per tablet, and that active ingredient makes up 40% of the total blend (with the rest being your excipient system — see how to make a direct-compression powder for how that ratio gets chosen), then:
200 mg ÷ 0.40 = 500 mg target tablet weight
The other 300 mg is excipient — diluent, binder, glidant, and lubricant. If you're using a pre-balanced blend like ReadyBlend, the active-to-excipient ratio is typically already specified for you within its 20–60% recommended range, so this step is mostly just arithmetic rather than formulation guesswork.
A quick sanity check worth running here: does 500 mg correspond to a reasonable tablet size for the die diameter you're planning to use? A very small die with a very high target weight will compress into an unusually thick tablet, which can affect both compression force and dwell time. If the numbers look mismatched, it's worth reconsidering die size before moving to the press.
Step 2: Translate Weight Into Fill Depth
Target weight is a number on paper — fill depth is the physical setting on the press that actually produces it. Every tablet press meters powder into the die cavity by volume, not weight, which means the same fill-depth setting will produce a different tablet weight for a denser powder than for a fluffier one. There's no universal formula for this conversion because it depends on your specific powder's bulk density, so it has to be calibrated on the press directly:
- Set an initial fill depth. Start with a middle-of-the-range setting on your press (see your press's manual — for example, how to use the TDP-0 for a manual bench press) as a starting point, not a final value.
- Press 10–20 tablets at that setting without changing anything between them.
- Weigh each tablet individually on a scale precise to at least 1 mg, and calculate the average.
- Compare the average to your target weight. If the average is above target, reduce the fill depth slightly; if below, increase it. Small adjustments — the equivalent of a partial turn on most fill-depth dials — go a long way, so change one variable at a time.
- Repeat with a fresh set of 10–20 tablets at the adjusted depth until the average lands on target.
Worked Example
Say you're formulating a 250 mg active ingredient dosed at 50% of the tablet:
- Target weight: 250 mg ÷ 0.50 = 500 mg
- First fill-depth trial produces an average of 540 mg across 15 tablets — 8% over target
- Fill depth is reduced slightly; a second trial produces an average of 505 mg — within a reasonable tolerance
- That fill-depth setting is locked in and used for the full production run, with periodic weight checks (see below) to confirm it holds
